Based on my own experience, I think it is important to have a responsible member of staff on site (or at least on call) at all times.
Guests can arrive and depart at any hour of day or night, and someone should be there to take care of them. Guests can also be irresponsible at times if there is no one to keep an eye on them, which can lead to problems with noise, theft, destruction of hostel property, non-paying visitors, etc. If an emergency situation arises (medical, fire, etc) you will want someone to be there who can handle it.
Whether you need someone to be there at night depends on the location and what the area is like, the type of guests that you want to attract, and the local laws.
