Twitter usage for a hostel would be more of an announcement channel where you can keep all the followers up to date with what's going on. Sometimes people misunderstand twitter and start expressing personal opinions on the official twitter account. That's half way to closing your twitter account :D
It can be very efficient specially if you use a software that allows you to write update once and then it updates twitter and facebook accounts.

Do you think Twitter can be used for hostels for marketing/promotion? Why would people follow a hostel in the first place?
Maybe it is better just to have a personal account and occasionally tweet about your hostel.

Many people will follow hostel Twitter accounts if they post interesting things and interacts with other users. I think the best use of Twitter isn't to expect direct bookings, but to network with travel bloggers, travel writers, media contacts, travel guides, tourism agencies, and other people that are good to know. You can often use Twitter to bypass regular channels.

I think it's better to have an account for the hostel rather than only a person, because that person may not stay with the business forever.
For hostels where multiple people are going to manage one Twitter account, there is free software called Cotweet. If only one person manages the Twitter account, Tweetdeck and the official app for mobile phones are good.
